In 2004 he played Nicholas Devereaux, the love interest of Anne Hathaway's character, in The Princess Diaries 2: Royal Engagement. He followed it with the romantic comedy Just My Luck (2006), opposite Lindsay Lohan, and the same year appeared in the action film Smokin' Aces. On stage he performed the one-man play The Atheist in New York in late 2006 and starred in the Los Angeles production of Fat Pig in 2007. In 2008 he played the vintner Bo Barrett in Bottle Shock.

He played the Coast Guard officer Bernard C. Webber in The Finest Hours (2016) and one of two brothers in the crime drama Hell or High Water, which premiered at the Cannes Film Festival in 2016.
His voice work extended to television: in July 2016 he received a Primetime Emmy Award nomination for Outstanding Character Voice-Over Performance for the animated series SuperMansion. That year he also appeared on Barbra Streisand's album Encore: Movie Partners Sing Broadway, performing a duet medley with her.
From 2017 he played Steve Trevor opposite Gal Gadot in Wonder Woman, a role he repeated in Wonder Woman 1984 (2020). He was also executive producer and a lead actor in the TNT series I Am the Night, which began airing in January 2019. In 2018 he played Dr. Alexander Murry in A Wrinkle in Time, starred as Robert the Bruce in the Netflix film Outlaw King and gave the voice of a version of Spider-Man in Spider-Man: Into the Spider-Verse. In 2022 he appeared in Olivia Wilde's Don't Worry Darling, and he acted and served as executive producer on The Contractor and on the espionage thriller All the Old Knives. He played Edgin Darvis in Dungeons & Dragons: Honor Among Thieves, released in March 2023, and voiced the antagonist King Magnifico in the Disney animated film Wish.
Pine made his directorial debut with Poolman, which he also wrote and starred in, and which was first shown at the Toronto International Film Festival in 2022 before its 2023 release.
